The HBsAb (Anti-HBs) – Quantitative Hormone Test Kit – ENZO is a quantitative enzyme-linked immunosorbent assay (ELISA) for the determination of antibodies to hepatitis B virus surface antigen (anti-HBs) in human serum or plasma. It is used for clinical purposes and assessing antibody response levels to HBsAg-vaccine.
The test is based on the principle of antigen-antibody binding. The microtiter wells of the test plate are coated with recombinant HBsAg. Patient serum or plasma is added to the wells together with a second HBsAg conjugated to horseradish peroxidase (HRP). If anti-HBs is present in the sample, the coated and conjugated antigens will bind to the two variable domains of the antibody and form an immune complex. After incubation, the unbound conjugates are washed away and the bound conjugates are detected by adding a substrate solution. The intensity of the color developed is proportional to the concentration of anti-HBs in the sample.
The results of the test are expressed in milli-international units per milliliter (mIU/mL). A negative result indicates that the patient is not immune to hepatitis B virus and should be vaccinated. A positive result indicates that the patient is immune to hepatitis B virus. An indeterminate result indicates that the patient has low levels of anti-HBs and may need to be revaccinated.
