Bacterial suspension that carry antigen can agglutinate on exposure to antibodies to salmonella organisms. Patients’ suffering from enteric fever would possess antibodies in their sera which can react and agglutinate serial doubling dilutions of killed, colored salmonella antigens in an agglutination test.
The main principle of widal test is that if homologous antibody is present in patients humor, it’ll react with various antigen within the reagent and provides visible clumping on the check card and agglutination within the tube. The antigens utilized in the check are “H” and “O” antigens of Salmonella typhosa and “H” antigen of S. Paratyphi. The paratyphoid “O” antigen are not used as they cross-react with typhoid “O” antigen due to the sharing of issue twelve. “O” antigen could be a somatic antigen and “H” antigen is a flagellar antigen.
